Terapia drożdżowa w leczeniu raka i jej wspomaganie MGN-3/Biobranem, arabinoksylanem z otrębów ryżowych POPARTE BADANIAMI

Apoptoza jest precyzyjnie zaprogramowanym mechanizmem do usuwania starych, uszkodzonych i nieprawidłowych komórek. W ciągu ostatnich dziesięciu lat badania pokazały obiecującą przyszłość terapii antynowotworowych opartych na apoptozie. Środki chemioterapeutyczne w sposób nieselektywny zabijają zarówno komórki rakowe jak i wiele zdrowych komórek. Dlatego też szczególnie ważne jest znalezienie środka, który byłby w stanie stymulować apoptozę konkretnie w komórkach nowotworowych przy minimalnych skutkach ubocznych. W ostatnim czasie pojawiło się zainteresowanie terapią nowotworową opartą na drobnoustrojach. Zastosowanie S. cerevisiae, drożdży piekarskich, jest oryginalnym podejściem do leczenia raka, które opiera się na założeniu, że komórki guza ulegają apoptozie przy fagocytozie drożdży. Nasze dane wskazują, że: 1) zabite termicznie drożdże wywołują apoptozę wielu ludzkich nowotworów (np., piersi, języka i jelita grubego) in vitro, 2) drożdże nie są toksyczne dla zdrowych komórek, 3) wywoływanie apoptozy jest specyficzne dla drożdży, 4) działanie apoptyczne drożdży na komórki nowotworowe jest wzmacniane MGN-3/Biobranem, modyfikatorem odpowiedzi biologicznej i, co bardzo ważne, 5) zjawisko apoptozy wywołanej drożdżami zostało zaobserwowane in vivo na modelu bezgrasicznych myszy bezwłosych (atymicznych myszy NUDE). Badania pod mikroskopem elektronowym próbek tkanek pobranych z guzów leczonych drożdżami pokazują wyraźnie, że komórki guzów fagocytują drożdże a następnie ulegają apoptozie. Mechanizm molekularny leżący u podłoża tego zjawiska jest obecnie badany w naszym laboratorium. Dalsze badanie w tym obszarze mogą pokazać implikacje dla leczenia nowotworów.
